An intracellular protein termed ERAB ( Endoplasmic reticulum associated Ab binding protein on chromosome X) has been cloned and linked with Ab neurotoxicity (1). ERAB has structural homology with short-chain alcohol dehydrogenases (hydroxysteroid dehydrogenases and acetyl-CoA reductases). Interestingly, ERAB localizes to endoplasmic reticulum despite the absence of a classical transmembrane sequence. However, in the presence of Ab, ERAB- Ab complex translates to the inner plasma membrane. Alzheimer patients have strong ERAB activity. Normal brains have virtually no ERAB. In addition, Ab neurotoxicity can be reduced by blocking ERAB and increased by its overexpression.

Human ERAB is an intracellular (endoplasmic reticulum), 264aa (Mouse ERAB 261aa), single polypeptide of approx. 27kD. It appears to ubiquitously expressed in normal brain and other tissue (1). Although, it is expressed at a higher levels in brains from Alzheimer patients. A 17 amino acid peptide sequence near the amino terminus of human ERAB was used to generate anti-ERAB. The peptide was coupled with KLH (carrier protein) and antibodies generated in rabbits.
ApplicationsSuitable for use in ELISA and Western Blot. Other applications not tested.
Recommended DilutionWestern Blot: 1:1000-1:5000
ELISA: 1:10,000-1:100,000 using 50-100ng of control peptide/well.

Storage and StabilityMay be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
