An intracellular protein termed ERAB ( Endoplasmic reticulum associated Ab binding protein on chromosome X) has been cloned and linked with Ab neurotoxicity (1). ERAB has structural homology with short-chain alcohol dehydrogenases (hydroxysteroid dehydrogenases and acetyl-CoA reductases). Interestingly, ERAB localizes to endoplasmic reticulum despite the absence of a classical transmembrane sequence. However, in the presence of Ab, ERAB- Ab complex translates to the inner plasma membrane. Alzheimer patients have strong ERAB activity. Normal brains have virtually no ERAB. In addition, Ab neurotoxicity can be reduced by blocking ERAB and increased by its overexpression.

ApplicationsSuitable for use in ELISA and Western Blot. Other applications not tested.
Recommended DilutionWestern Blot: 1:1000-1:5000 for neat serum and 1-10ug/ml for affinity pure using ECL. An antibody made to the ERAB11 epitope has detected ~ 27kD protein in the brain.
ELISA: 1:10,000-1:100,000 using 50-100ng of control peptide/well.
Optimal dilutions to be determined by the researcher.

Storage and StabilityMay be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Freeze at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
