| NO, produced by nitric oxide synthases (NOS), is a critical signaling molecule involved in many physiological processes including muscle contraction, vasodilation, neuronal transmission, and immune responses. Some of these effects are mediated through reaction with and activation of guanylate cyclase (GC), resulting in increased cGMP production. With a half-life of seconds, NO readily diffuses through the cytosol and cell membrane, allowing it to act inside or adjacent to the cell in which it was produced. | | | Catalog # | E3218 | | Applications | Suitable for use in Western Blot. Other applications have not been tested. | | Recommended Dilution | Western Blot: 1:1000 detects recombinant eNOS phosphorylated by PKA and endogenous eNOS from VEGF treated HUVAC cell lysates. HUVAC and VEGF treated HUVAC cell lysate were resolved by electrophoresis, transferred to nitrocellulose and probed with E3218. Proteins were visualized using a goat anti-rabbit secondary antibody conjugated to HRP and a chemiluminescence detection system. | | Optimal dilutions to be determined by the researcher. | | Recommended Control | Brain tissue, VEGF treated HUVAC cell lysate | | Storage and Stability | May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. | | CAS Number | n/a | | Clone Type | Polyclonal | | Isotype | IgG | | Host | Rabbit | | Source | Human | | Concentration | Not determined | | Form | Supplied as a liquid in buffer, 0.05% sodium azide, 30% glycerol. | | Purity | Serum | | Immunogen | Synthetic peptide corresponding to aa1172-1183, C-RIRTQ[pS]FSLQER, of human eNOS (endothelial nitric oxide synthase) with an N-terminal cysteine added (KLH). | | Specificity | Recognizes phosphorylated human eNOS, Mr 132kD. Additional unknown bands at 50 and 64kD Species sequence homology: rat, mouse, bovine, porcine and canine. | | | Important Note | This product as supplied is intended for research use only, not for use in human, therapeutic or diagnostic applications without the expressed written authorization of United States Biological. |
