NO, produced by nitric oxide synthases (NOS), is a critical signaling molecule involved in many physiological processes including muscle contraction, vasodilation, neuronal transmission, and immune responses. Some of these effects are mediated through reaction with and activation of guanylate cyclase (GC), resulting in increased cGMP production. With a half-life of seconds, NO readily diffuses through the cytosol and cell membrane, allowing it to act inside or adjacent to the cell in which it was produced.

ApplicationsSuitable for use in Western Blot. Other applications have not been tested.
Recommended DilutionWestern Blot: 1:1000 detects recombinant eNOS phosphorylated by PKA and endogenous eNOS from VEGF treated HUVAC cell lysates. HUVAC and VEGF treated HUVAC cell lysate were resolved by electrophoresis, transferred to nitrocellulose and probed with E3218. Proteins were visualized using a goat anti-rabbit secondary antibody conjugated to HRP and a chemiluminescence detection system.
Optimal dilutions to be determined by the researcher.
Recommended ControlBrain tissue, VEGF treated HUVAC cell lysate
Storage and StabilityMay be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
