EGFR is a type I recpetor tyrosine kinase with sequence homology to erb-B 1, 2, 3, 4, or HER 1, 2, 3, 4. It binds to epidermal growth factor, transforming growth factor-a, heparin binding EGF, amphiregulin, betacellulin, and epiregulin. EGFR is overexpressed in tumors of breast, brain, bladder, lung, gastric, head and neck, esophagus, cervix, vulva, ovary and endometrium. It is predominantly present in squamous cell carcinomas.

ApplicationsSuitable for use in Flow Cytometry, Immunofluorescence, Western Blot, Immunoprecipitation and Immunohistochemistry. Other applications have not been tested.
Recommended DilutionWestern Blot: 0.5-1ug/ml for 2 hours at RT
Immunoprecipitation (Native only): 2ug/mg protein lysate using Protein G.
Immunohistochemistry (Formalin/paraffin): 2-4ug/ml for 30 minutes at RT. Staining of formalin-fixed tissues requires digestion with protease XXV at 1mg/ml PBS for 5 minutes at 37°C.
Optimal dilutions to be determined by the researcher.
Positive ControlA431 cells. Placenta or squamous cell carcinoma.
Storage and StabilityMay be stored at 4°C for short-term only. For long-term storage, store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

SpecificityRecognizes the 170kD (wild type) and 145kD (VIII variant) forms of human EGF receptor. The epitope maps to the extracellular domain of EGFR. Does not react with mouse or rat.
