Estrogen regulates various physiological functions upon binding to its specific receptors. Estrogen receptors (ERs) are members of the nuclear receptor superfamily of ligand-dependent transcription factors that regulate the expression of target gene. ERa and ERb, which are encoded by different genes, have similar overall structure with at least three functional domains in common: the ligand-binding domain located at the C-terminal half of the proteins, the DNA-binding domain located centrally, and the N-terminus. Both ERa and ERb are co- expressed in the same cells such as brain, pituitary, and mammary gland. The co-expression of the two isoforms is a key determinant in the cellular responses to estrogen.
