Clone Host Grade Applications
Polyclonal Rabbit Affinity Purified E B IC IF
Estrogen receptor alpha (ER alpha), a member of the steroid receptor superfamily, contains highly conserved DNA binding
(DBD) and ligand binding domains (LBD) (1). Through its estrogen-independent and estrogen- dependent activation domains (AF-1 and AF-2, respectively), ER alpha regulates transcription by recruiting coactivator proteins and interacting with general transcriptional machinery (2). Phosphorylation provides an important mechanism to regulate ER alpha activity (3,4). ER alpha is phosphorylated on multiple sites (5). Serines 104 and 106 are located in the N-terminal transcription activation function domain AF-1, and phosphorylation of Ser104/106 plays an important role in regulating ER alpha activity (5).

ApplicationsSuitable for use in ELISA, Western Blot, Immunocytochemistry and Immunofluorescence.
Other applications not tested.
Recommended DilutionWestern Blot: 1:1000
Immunofluorescence: 1:50
Optimal dilutions to be determined by the researcher.
Storage and StabilityMay be stored at 4°C for short-term only. For long-term storage, store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

FormSupplied as liquid in 10mM HEPES, pH 7.5, 150mM sodium chloride, 0.1mg/ml BSA, 50% glycerol. No preservative added.
PurityPurified by Protein A affinity chromatography.
ImmunogenSynthetic phospho-Ser104/106 peptide (KLH coupled) corresponding to residues surrounding Ser104/106 of human ER alpha.
SpecificityDetects overexpressed levels of Ser104/106 phosphorylated ER alpha. Detects ER alpha only when phosphorylated at Ser104/106. It does not crossreact with phosphorylated ER isoform beta. Species Crossreactivity: Predicted to react with the specified phosphorylated motif from mouse and equine based on sequence homology.
