Human von Willebrand factor (factor VIII RAg) is a 270kD multimeric plasma gylcoprotein. It mediates platelet adhesion to injured vessel walls and serves as a carrier and stabilizer for coagulation factor VIII. The von Willebrand factor has functional binding domains to platelet glycoprotein Ib, glycoprotein IIb/IIIa, collagen and heparin. The factor is synthesized by endothelial cells and is also present in platelets and megakaryocytes.
ApplicationsSuitable for use in Immunofluorescence and Immunohistochemistry. Other applications not tested.
Recommended DilutionImmunohistochemistry (Paraffin): 1:200-1:400 using indirect Immunoflourescence. Enzymatically pre-digested with 0.1% pronase, formalin fixed. Specifically stains endothelial cells.
Immunohistochemistry (Paraffin): 1:1000 using indirect immunoperoxidase. Enzymatically pre-digested with 0.1% pronase, formalin fixed and developed with Biotin/Avidin. Specifically stains blood vessels.
Optimal dilutions to be determined by the researcher.
Positive ControlFormalin-fixed, paraffin sections of human tongue
Storage and StabilityMay be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

SpecificityRecognizes the human Factor VIII Related Antigen. Specifically reacts with the cytoplasm of human endothelial cells from normal and neoplastic blood and from lymphatic vessels. Also reacts with human endocardium, platelets and megakaryocytes. Factor VIII R:Ag endocytosed by other cells may also react.
