| This product consists of a pair of control extracts made from NIH3T3 cells. The cells are stimulated (+PDGF), or not (control) with Platelet Derived Growth Factor. Cells were extracted with modified RIPA buffer (50mM Tris, 150mM NaCl, 1% Triton-X 100, 0.5% deoxycholate, pH 7.3). The extracts can be used as controls for testing antibodies. | | | Catalog # | F4209-01 | | Applications | Suitable for use in Western Blot (with phosphorylation site specific antibodies (PSSAs) to ERK1&2 [pTpY185/187] and PLCg-1 [pY783]). Other applications not tested. | | Western Blot Conditions | Prior to loading, heat the extract sample for 90 seconds at 95-100°C, then spin in a microfuge for 5 seconds at RT. We recommend using 20ml of the extract per lane for SDS-PAGE, and transferring onto a nitrocellulose membrane. At 20ml per lane, the amount provided is sufficient for four lanes on a standard mini-gel. The optimal amount of extract used should be determined empirically for each specific primary antibody tested. | | Storage and Stability | May be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Freeze at -20°C or colder. Aliquots are stable for at least 6 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
