| FHOD1, a predominantly cytoplasmic protein, is a 1164aa containing member of formin:diaphanous family of proteins that contains a FH1 (formin homology 1) domain, a FH2 (formin homology 2) domain and a GBD/FH3 (Rho GTPase-binding:formin homology 3) domain. FHOD1 self-associates via the FH2 domain and binds to F-actin via its N-terminus and also to the cytoplasmic domain of CD21 via its C-terminus. Function of FHOD1 include the assembly of F-actin structures, such as stress fibers, and depends on the Rho-ROCK cascade for its activity. Reports suggest the role of FHOD1 in the coordination of microtubules with actin fibers and also in cell elongation. Endogenous FHOD1 interacts directly with Rac1 in a guanine nucleotide-independent manner and thus activates transcription from the serum response element.
