The enzyme beta galactosidase is a component of assays used frequently in genetics, molecular biology (see X-gal) for a blue white screen, and other life sciences. IPTG induces production of b-galactosidase by binding and inhibiting the lac repressor. Since it is highly expressed and accumulated in lysosomes in senescent cells, it is used as a senescence biomarker both in vivo and in vitro in qualitative and quantitative assays, despite its limitations.

ApplicationsSuitable for use in ELISA, Western Blot, Dot Blot, Immunofluorescence Microscopy, Immunohistochemistry, Immunoprecipitation and Conjugation. Other applications not tested.
Recommended DilutionsELISA: 1:10,000
Western Blot: 1:5000-1:10,000
Immunohistochemistry (4% paraformaldehyde fixed tissues, frozen, paraffin): 1:1500 detected b-galactosidase in adult rat spinal cord tissue after infection with helper-dependent adenovirus expressing lacZ. Tissue was perfused with 4% paraformaldehyde and cryostat-cut (35um) to produce free-floating sections.
Optimal dilutions to be determined by the researcher.
Storage and StabilityMay be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

SpecificityRecognizes b-Galactosidase. Assay by IEP resulted in a single precipitin arc against anti-rabbit serum as well as purified and partially purified b-Galactosidase (E. coli). Crossreactivity against b-Galactosidase from other tissues and species may occur.
