| Most mammalian cells transport glucose through a family of membrane proteins known as glucose transporters (Glut or SLC2A family). Molecular cloning of these glucose transporters has identified a family of closely related genes that encodes at least 9 proteins (Glut-1 to Glut-14, MW 40-60 kD). Individual member of this family have identical predicted secondary structures with 12 transmembrane domains. Both N and C-termini are predicted to be cytoplasmic. There is a large extracellualr domain between TM1-TM2 and a cytoplasmic domain between TM6-TM7. Most differences in sequence homology exist within the four hydrophilic domains that may play a role in tissue-specific targeting.Glut isoforms differ in their tissue expression, substrate specificity and kinetic characteristics. Glut-3 is the main transporter in neurons, whereas Glut-4 is primarily expressed in muscle and adipose tissue and regulated by insulin. | | | Catalog # | G3900-26 | | Applications | Suitable for use in ELISA and Western Blot. Other applications not tested. | | Recommended Dilution | Western Blot: 1-10ug/ml using ECL. Major band at ~45kD. | | ELISA: 0.5-1ug/ml. Coat ELISA plates at 1ug/ml (antigen) | | Optimal dilutions to be determined by the researcher. | | Storage and Stability | May be stored at 4°C for short-term only. Store long-term at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. | | CAS Number | n/a | | Clone Type | Polyclonal | | Isotype | IgG | | Host | Rabbit | | Source | Mouse | | Concentration | ~1mg/ml | | Form | Supplied as a liquid in 10mM Tris, pH 7.5, 0.2% BSA, 0.05% sodium azide, 40% glycerol. | | Purity | Purified by immunoaffinity chromatography. | | Immunogen | A 12 aa C-terminal peptide sequence near the C-terminus of mouse glucose transporter-3 (Glut-3). A cysteine has been added at the NH2 terminus for coupling to KLH. | | Specificity | Recognizes Glut-3. The mouse Glut-3 peptide sequence used for antibody production is 100% identical in rat. This antibody is specific for mouse and rat. Does not recognize human Glut-3 because of significant interspecies differences in C-terminal Glut-3. | | | Important Note | This product as supplied is intended for research use only, not for use in human, therapeutic or diagnostic applications without the expressed written authorization of United States Biological. |
