ApplicationsSuitable for use in ELISA, Western Blotting. Other applications not tested.
Recommended DilutionWestern Blot: 1:1000-1:5000 for neat serum and 1-10ug/ml for affinity pure antibody using ECL. An antibody raised to the rat C-terminal 13aa of glut-5 has been shown to react with a protein band of about 60kD in the rat jejunum membranes that can be competed with the control peptide (2, 3). The MW. Of Glut 5 is larger than that reported for human Glut 5 (50-55kD) (4). As opposed to human tissues, rat Glut-5 mRNA has not been detected in rat testis, adipose tissues, and skeletal muscle (2, 4). In rat kidney, a protein of about 42kD was detected (6). Differences in MW. Of Glut-3 in various tissues has also been noted.
ELISA 1:10,000-50,000 for neat serum and 0.5-1ug/ml for affinity pure. Control peptide can be used to coat ELISA plates at 1ug/ml.
Optimal dilutions to be determined by the researcher.
Storage and StabilityMay be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Freeze at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

SpecificityHuman GT52 sequences shows about 61% homology with rat Glut-5 (8/13 AA). For rat Glut-5, we suggest the use of antibodies that are made to a peptide from rat Glut-5. Antibody crossreactivity in various species is not established. Control peptide, because of its low mol size (<3kD), is not recommended for Western. It should be used in ELISA, Dot Blot and for antibody blocking to confirm specificity of antibodies
