HSP27 is identical to the estrogen-induced p29 and 24K protein. About 50% of breast carcinomas are positive for HSP27 especially those that are also positive for estrogen and/or progesterone receptor. HSP27 has also been implicated in drug resistance in cancer cells.

Synthesis of heat shock and stress-related proteins is stimulated by cellular stress, thus enabling the cell to recover from and survive stressful conditions. The mechanisms behind this recovery process are as yet unknown. Due to their role in cellular repair and the fact that they are overexpressed in many tumors, heat shock proteins are now an important subject of tumor marker research. Hsp27 was recently found identical to the estrogen-induced p29 and 24K proteins (1). Production of hsp27 has been shown to be triggered by both heat shock (in estrogen-independant as well as estrogen-dependent cell lines) and by estrogen administration. This effect was reversed by administration of antiestrogens such as Tamoxifen (2).
Hsp27 protein is found in both normal and neoplastic tissues, in varying amounts. High to moderate espression was found primarily in normal human uterus (endometrium), vagina (epithelium), Fallopian tubes (ciliated cells), uterine cervix (epithelium), placenta (decidual cells), and skin (strata spinosum and ducts of sweat glands) (3-4). Low levels of hsp27 were detected in mammary glands (ductal cells in some lobules) and smooth & striated muscle (3-4).
Hsp27 has been evaluated for its prognostic applications in breast and other tumors. To date, the data suporting its use as a prognistic marker in breast cancer are contradictory. Several studies (5-8) indicate that increased levels of expession may be used as a marker of good prognosis and increased likelihood of response to hormonal therapy. Other researchers (9-11) disprove these claims, nding that increased espression is asociated with markers of reduced survival and poor prognosis.
The most promising research with hsp27 has been in the area of drug-resistance. Studies show that, while simultaneous administration of hypothermia and chemotherapy shows reduced effectiveness (12-13). Additonal research by Oesterreich, et al. (14) shows selective resistance to doxorubicin in breast cancer cells after heat shock. In this study, transfection experiments showd that MDA-MB-231 breast cancer cells, which normally express low levels of protein, gained a three-fold resistance to doxorubicin after transfection with a full-length construct. Similarly, when an MCF-7 subline displaying ampli ed expression levels was transfected with an antisense construct, resistance to doxorubicin dropped 3-fold.
ApplicationsSuitable for use in Western Blot and Immunohistochemistry. Other applications have not been tested.
Recommended DilutionWestern Blot: 1-2ug/ml for 2 hours at RT.
Immunohistochemistry (Formalin/paraffin): 1-2ug/ml for 30 minutes at RT. Staining of formalin- xed tissues requires boiling tissue sections in10mM citrate buffer, pH 6.0 for 10-20 min., followed by cooling at RT for 20 min.
Optimal dilutions to be determined by the researcher.

Storage and StabilityMay be stored at 4°C for short-term only. For long-term storage, store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
