Clone Host Grade Applications
Polyclonal Rabbit Affinity Purified E B IH IC
Heat shock protein (HSP) 27 is one of the small HSPs that are constitutively expressed at different levels in various cell types and tissues. Like other small heat shock proteins, HSP27 is regulated at both the transcriptional and posttranslational levels (1). In response to stress, the expression level of HSP27 increases several-fold in order to confer cellular resistance to the adverse environmental change. HSP27 is also phosphorylated at serines 15, 78 and 82 by MAPKAP kinase 2 as the result of the activation of the p38 MAP kinase pathway (2,3). Phosphorylation of HSP27 causes a change in its tertiary structure, which shifts from large homotypic multimers to dimers and monomers (4). It has been illustrated that phosphorylation and increased concentration of HSP27 modulate actin polymerization and reorganization (5,6).

ApplicationsSuitable for use in Western Blot, Immunohistochemistry (Paraffin) and Immunocytochemistry (ABC). Other applications have not been tested.
Recommended DilutionWestern Blot: 1:1000. Incubate membrane with diluted antibody in 5% BSA, 1XTBS, 0.1% Tween-20 at 4°C with gentle shaking, overnight.
Immunohistochemistry (Paraffin): 1:50
Immunocytochemistry (ABC): 1:500
Optimal dilutions to be determined by the researcher.
Storage and StabilityMay be stored at 4°C for short-term only. For long-term storage, store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
