| Proteoglycans are macromolecules consisting of a variety of core proteins with covalently attached one or several polysaccharide chains of the glycosaminoglycan type (heparan sulfate, heparin, chondroitin sulfate, dermatan sulfate or keratan sulfate). At least two forms of basement membrane heparan sulfate proteoglycan (HSPG) have been identified. One with a large core protein (> 400kD) and one with a small core protein (30kD). The large HSPG is probably the most abundant basement membrane proteoglycan. It is located predominantly in the lamina lucida, where it forms clustered aggregates and interacts with other basement membrane components to form the matrix. In addition, it also plays a critical role in attachment of cells to the basal membrane via integrin receptors. | | | Catalog # | H1890-90F | | Applications | Suitable for use in Immunofluorescence and Immunohistochemistry. Other applications not tested. | | Recommended Dilution | Immunohistochemistry: 1-2ug/ml | | Note: Staining of acid-alcohol fixed tissues requires unmasking with 15,000ul/ml of bovine testicular hyaluronidase in PBS, pH 7.4 for 30 minutes at 37ºC. | | Optimal dilutions to be determined by the researcher. | | Positive Control | Tonsil | | Storage and Stability | May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. | | CAS Number | n/a | | Clone Type | Monoclonal | | Isotype | IgG2a | | Clone No | 8L159 | | Host | Rat | | Source | Mouse | | Concentration | ~0.2mg/ml | | Form | Supplied as a liquid in 10mM PBS, pH7.4, 0.2% BSA, 0.09% sodium azide. | | Purity | Purified by Protein G affinity chromatography. | | Immunogen | Murine EHS laminin preparation. Cellular Localization: Basement membrane. | | Specificity | Recognizes mouse domain IV of the core protein of the large heparan sulfate proteoglycan. The reactivity is independent of the galactosaminoglycan moieties. Therefore, the epitope is not sensitive to heparitinase treatment. Species Crossreactivity: Human, porcine, bovine | | | Important Note | This product as supplied is intended for research use only, not for use in human, therapeutic or diagnostic applications without the expressed written authorization of United States Biological. |
