| Perlecan is an extracellular matrix proteoglycan. It has a large core protein of 400-450kD and is often produced with heparan sulfate side chains. Perlecan is found in basement membranes where it contributes to the permeability charateristics, serves as a substrate for vascular cells and binds growth factors involved in vascular remodelling (2). | | | Catalog # | H1890-91 | | Applications | Suitable for use in Immunocytochemistry. Other applications have not been tested. | | Recommended Dilution | Immunocytochemistry: 10ug/ml on primary cultures of hippocampal neurons, fixed with 4% paraformaldehyde in phosphate buffer containing 4% sucrose. The neurons were subsequently permeabilized with 0.25% Triton X-100 for 5 minutes at RT. The fixed and permeabilized cells were blocked with normal donkey serum in Tris buffered saline (TBS) containing 10% Tween. The primary antibody was detected using a FITC-conjugated goat anti-rat secondary antibody. | | Optimal dilutions to be determined by the researcher. | | Hybridoma | Mouse-rat hybridoma A7L6 (X63.Ag.8 653 mouse myeloma x Fisher rat splenocytes). Propagated in cell culture. | | Storage and Stability | May be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, add sterile glycerol (40-50%), aliquot and store at -20°C or colder. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
