Clone Host Grade Applications
Polyclonal Rabbit Serum B
Histones are relatively small proteins that contain a high proportion of positively charged amino acids, e.g., arginine and lysine. This positive charge allows histones to bind tightly to the negatively charged DNA. There are five basic types of histone molecules called H1, H2A, H2B, H3, and H4. Histones are abundantly present in virtually all eukaryotic cells.
Catalog #H5110-08S
ApplicationsSuitable for use in Western Blot. Other applications not tested.
Recommended DIlutionsWestern Blot: 1:2000-1:10,000 detects phosphorylated Histone H2A,H4 (Ser1) in 10ug of acid-extracted proteins from colcemid-arrested HeLa cells. Acid-extracted proteins from untreated HeLa cells and from colcemid-arrested HeLa cells were resolved by electrophoresis, transferred to nitrocellulose and probed with H5110-08S (1:5000). Proteins were visualized using a goat anti-rabbit secondary antibody conjugated to HRP and a chemiluminescence detection system.
Optimal dilutions to be determined by the researcher.
Recommended Secondary AntibodiesI1904-39: IgG, X-Adsorbed (HRP) Pab Gt xRb
I1904-40A: IgG, H&L, X-Adsorbed (HRP) Pab Gt xRb
I1904-46Q: IgG, H&L (HRP) Pab Gt xRb

ImmunogenSynthetic peptide corresponding to aa1-9, [pS]GRGKQGGK-C, of human histone H2A with a C-terminal cysteine added to facilitate conjugation. Conjugated to KLH. The immunizing sequence has 8/9 amino acid identity with Histone H4.
SpecificityRecognizes phosphorylated human Histones H2A and H4, Mr 16kD and 10kD respectively. Broad species crossreactivity expected due to sequence conservation.
Important NoteThis product as supplied is intended for research use only, not for use in human, therapeutic or diagnostic applications without the expressed written authorization of United States Biological.
