Histone 2B is the core component of nucleosome. It is involved in nucleosome wrapping and the compaction of DNA into chromatin, limiting DNA accessibility. Histones thereby play a central role in transcription regulation, DNA repair, DNA replication, and chromosomal stability. DNA accessibility is regulated via a complex system of nucleosome remodelling and post-translational modification of histones, also called histone code.

ApplicationsSuitable for use in ELISA, Western Blot, Immunocytochemistry and Immunohistochemistry. Other applications not tested.
Recommended DilutionWestern Blot: 1:1000-1:10,000. Detected phospho-Histone H2B in HL-60 cells treated with 20ug/ml VP16 for 18 hours. A 1:250-1:2000 dilution also detected phospho-Histone H2B acid extracted proteins from colcemid-arrested HeLa cells.
Immunocytochemistry: 1:5000 selectively immunostained pycnotic apoptotic HL-60 cells and TUNEL positive HeLa cells.
Immunohistochemistry: 1:5000 immunostained deparaffinated Xenopus tail sections.
Note: Use of this antibody at dilutions which are too low will result in non-specific detection of Histone H2B and other unknown proteins in immunoblots of acid extracts from untreated cells.
Optimal dilutions to be determined by the researcher.
Storage and StabilityMay be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
