ApplicationsSuitable for use in Western Blot, Immunocytochemistry and Peptide inhibition. Other applications not tested.
Recommended DilutionWestern Blot: 0.05-0.4ug/ml detects phosphorylated histone H3 in acid extracted proteins from mitotic HeLa cells treated with colcemid, but not unmodified recombinant Histone H3. Acid extract from untreated and colcemid treated HeLa cells, along with recombinant Histone H3 were resolved by electrophoresis, transferred to nitrocellulose and probed with anti-phospho-Histone H3 (Ser10) (0.1ug/ml). Proteins were visualized using a goat anti-mouse secondary antibody conjugated to HRP and a chemiluminescence detection system.
Peptide Inhibition Analysis Detection of histone H3 in immunoblots of colcemid treated HeLa acid extracts by anti-phospho-Histone H3 (Ser10) was diminished by 10uM of histone H3 peptide containing phospho-serine 10, but not by peptides containing phospho-serine 28 or an unmodified histone H3 sequence
Immunocytochemistry 0.2ug/ml showed positive chromosome immunostaining for mitotic A431 and HeLa cells fixed with 95% ethanol and 5% acetic acid and permeabilized with 0.1% Triton® X-100.
Optimal dilutions to be determined by the researcher.
Storage and StabilityMay be stored at 4°C for short-term only. For long-term storage, store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
