| C-ADPSPQNSTADTSK, from the internal region of human HMGCR. | | | Control peptide corresponding to polyclonal antibody, Catalog#H6201-01A, goat x human. | | | Catalog # | H6201-01A-P | | HMG-CoA reductase is the rate-limiting enzyme for cholesterol synthesis and is regulated via a negative feedback mechanism mediated by sterols and non-sterol metabolites derived from mevalonate, the product of the reaction catalyzed by reductase. Normally in mammalian cells this enzyme is suppressed by cholesterol derived from the internalization and degradation of low density lipoprotein (LDL) via the LDL receptor. Competitive inhibitors of the reductase induce the expression of LDL receptors in the liver, which in turn increases the catabolism of plasma LDL and lowers the plasma concentration of cholesterol, an important determinant of atherosclerosis. Alternatively spliced transcript variants encoding different isoforms have been found for this gene. | | Applications | Suitable for use in ELISA. Other applications not tested. | | Recommended Dilutions | Optimal dilutions to be determined by the researcher. | | Storage and Stability | May be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and store at -20°C. Aliquots are stable for at least 12 monthsat -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. | | Peptide Blocking (see corresponding antibody H6201-01A) | Antibodies are typically supplied at 0.5mg/ml and peptides as a 100ul pellet. When peptides are reconstituted in 200ul water, the concentration would be also 0.5mg/ml. To start, the best ratio would be 1:1 (which means molar excess of peptides relative to antibodies when identical volumes are mixed). Mix equal volumes of peptide and antibody at the required dilution and leave at ambient temperature. It is best is to have two identical blots, to be incubated with equal amount of antibodies, but one with the antibodies pre-adsorbed to the peptide for 20min.
