| NFkB exists in the cytoplasm as a complex with a 38kDa inhibitor protein, IkB (MAD-3). Activation of NFkB by different stimuli, including TNFa and PMA, requires the dissociation of the inhibitor protein. Dissociation is regulated by the phosphorylation of IkB on serine residues 32 and 36 which targets IkB for ubiquinitation and proteasome mediated degradation.
