Type I insulin-like growth factor receptor (IGF-IR) is a transmembrane tyrosine kinase. It is widely expressed among many cell types in fetal and postnatal tissues and many cell lines. Upon binding of its ligands, IGF-I and IGF-II, autophosphorylation of receptors occurs. The triple tyrosine cluster (Tyr1131, Tyr1135 and Tyr1136) within the kinase domain is the earliest major site of autophosphorylation. Phosphorylation of these three tyrosine residues is necessary for activation of kinases. Insulin receptors (IRs) share significant similarity with IGF-I receptors in structure and function. There is also an equivalent triple tyrosine cluster (Tyr1146, Tyr1150 and Tyr1151) in its activation loop of the kinase domain. Tyrosine autophosphorylation of the receptors is one of the earliest cellular responses to insulin stimulation. The autophosphorylation begins by phosphorylation of Tyr1146 and either Tyr1150 or Tyr1151. The full activation of receptor kinase activity depends on the triple tyrosine phosphorylation.
ApplicationUse at 250uM final concentration as the substrate for in vitro IGF-1R kinase assays.
Protein Kinase Assay: At 250uM, this lot was successfully phosphorylated by IGF-1R ( 1-958), active. Assay results show a > 20-fold increase in phosphate transferred to the peptide over background using 500ng of enzyme.
Storage and StabilityLyophilized powder may be stored at 4°C for short-term only. Stable for 12 months at -20°C. Reconstitute to nominal volume (see reconstitution instructions for peptides) and store at -20°C. For maximum recovery of product, centrifuge the original vial prior to removing the cap. Further dilutions can be made in assay buffer.
