Clone Host Grade Applications
Polyclonal Rabbit Affinity Purified B IP
Integrin a4 (also called CD49d) is a ~150kD protein that possesses a large extracellular domain involved in ligand binding, a single transmembrane domain, and an intracellular regulatory domain possessing multiple sites for phosphorylation. Integrin a4 forms heterodimers with integrins b1 and b7. Integrin a4 is expressed on leukocytes and leukocyte precursors, neural crest cells, and developing skeletal muscles and is essential for embryogenesis, hematopoiesis, and immune responses. The presence of integrin a4 promotes cell migration and inhibits cell spreading and mcontractility. Integrin a4 function has been implicated in the pathogenesis of multiple diseases including asthma, rheumatoid arthritis, Crohn’s disease, ulcerative colitis, hepatitis C, and multiple sclerosis, and therefore, modulation of integrin a4 function has become an important target for drug discovery. Phosphorylation of the serine 988 residue on the integrin a4 cytoplasmic tail inhibits its interaction with the focal adhesion complex-associated protein paxillin, and modulates its effects on cell motility and spreading.
Catalog #I7661-34A
ApplicationsSuitable for use in Western Blot and immunoprecipitation applications. Other applications have not been tested.
Recommended DilutionWestern Blot: 1:500–1:1000. A 1:1,000 dilution provides 100ml working solution, which at 10ml/blot allows 10 blots to be performed.
Optimal dilution determined by the researcher.
Positive ControlJurkat T cells transfected with full-length, wild-type or S988A mutant integrin a4.
Storage and StabilityMay be stored at 4°C for short-term only. For long-term storage, store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
Clone TypePolyclonal
IsotypeIgG
HostRabbit
SourceHuman
ConcentrationNot determined.
FormSupplied as a liquid in PBS, pH 7.2, 0.05% sodium azide.
PurityPurified by immunoaffinity chromatography.
ImmunogenSynthetic phosphopeptide derived from a region of human integrin a4 that contains serine 988.
SpecificityRecognizes human integrin a4 when phosphorylated at Ser988.. The peptide corresponding to integrin a4 [pS988] blocks the antibody signal, and that the antibody does not react with the S988A mutant, thereby demonstrating its specificity.
