IL-33 is a member of the IL-1 cytokine family and is constitutively expressed in smooth muscle cells and the airway epithelium. IL-33 signals through receptors, IL-1RL1 and IL-1RAP, to induce type 2 cytokine production in polarized Th2 cells.
Applications
Suitable for use in Western Blot, Flow Cytometry, Immunocytochemistry, ELISA, Immunohistochemistry and Neutralization. Other applications not tested.
Recommended Dilutions
Western Blot: 0.4ug/ml using HEK293T human embryonic kidney cell line transfected with full length mouse IL-33. Flow Cytometry (Intracellular): 2.5ug/10e6 mouse splenocytes treated with PMA and Ca2+ ionomycin, fixed with paraformaldehyde and permeabilized with saponin. Immunocytochemistry: 5-15ug/ml using Immersion fixed bEnd.3 mouse endothelioma cell line Immunohistochemistry: 5-15ug/ml using perfusion fixed frozen sections of mouse spleen ELISA (Capture): 0.2-0.8ug/ml Neutralization: 10-50ng/ml in the presence of 0.25ng/ml of recombinant mouse IL-33. Optimal dilutions to be determined by the researcher.
Storage and Stability
Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
Immunogen
Recombinant protein corresponding to Ser109-Ile266 from mouse IL-33 expressed in E. coli.
Form
Supplied as a lyophilized powder from PBS, pH 7.4, 5% trehalose. Reconstitute with 500ul sterile PBS.
Purity
Purified by Immunoaffinity chromatography.
Specificity
Recognizes mouse IL-33. In sandwich immunoassays, less than 2% crossreactivity is observed with recombinant human IL-33.
