| Interleukin-4 (IL-4) is a potent lymphoid cell growth factor which stimulates the growth and survivability of certain B cells and T cells. IL-4 is a pleiotropic cytokine that is produced by activated T cells, mast cells and basophils. IL-4 elicits many different biological responses, but has two dominant functions. The first is regulating differentiation of naive CD4+ T cell to the TH2 type. TH2 cells produce IL-4, IL-5, IL-10 and IL-13, which tend to favor a humoral immune response while suppressing a cell-mediated immune response controlled bu TH1 cells. The second is regulating IgE and IgG1 production by B cells. Recombinant IL-4 lacks all N-linked oligosaccharides to molecules highly overglycosylated, but all are fully biologically active. IL-4’s biological activity is species-specific. | | | Catalog # | I8426 | | Recombinant Human IL-4 produced in E. coli is a single, non-glycosylated polypeptide chain containing 130 amino acids and having a molecular mass of 15000D. | | Biological Activity | Human IL-4 is fully biologically active when compared to standards. The ED50 as determined by the dose-dependent stimulation of TF-1 cells is 0.2 ng/ml, corresponding to a specific activity of 5x10e6 units/mg. | | Storage and stability | Lyophilized powder may be stored at 4°C for short-term only. Reconstitute to nominal volume by adding sterile ddH2O, 0.1% HSA or BSA, aliquot and store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay bufferbuffer. | | Amino acid sequence: | | The sequence of the first five N-terminal amino acids was determined and was found to be Met-His-Lys-Cys-Asp. | | Dimers and aggregates | 1% as determined by silver-stained SDS-PAGE gel analysis. | | Biological Activity | Recombinant Human Interleukin-4 is fully biologically active when compared to standard. The ED50 as determined by the dose-dependent stimulation of TF-1 cells is less then 0.2ng/ml, corresponding to a Specific Activity of 5x10 IU/mg. | | Protein | Protein quantitation was carried out by two independent methods: | | 1. UV spectroscopy at 280nm using the absorbency value of 0.594 as the extinction coefficient for a 0.1% (1mg/ml) solution. This value is calculated by the PC GENE computer analysis program of protein sequences (IntelliGenetics). | | 2. Analysis by RP-HPLC, using a standard solution of IL-4 as a Reference Standard. | | Molecular Weight | 14.9kD | | Source | Recombinant, Human (E. coli). | | Purity | 98% by SDS-PAGE and HPLC | | Form | Supplied as a sterile filtered, lyophilized protein. No additives. | | | Reconstitute in sterile dH2O to a concentration of 0.1-1mg/ml. This solution can then be diluted into other aqueous solutions. | | | Important Note | This product as supplied is intended for research use only, not for use in human, therapeutic or diagnostic applications without the expressed written authorization of United States Biological. |
