Interleukin-4 (IL-4) is a potent lymphoid cell growth factor which stimulates the growth and survivability of certain B cells and T cells. IL-4 is a pleiotropic cytokine that is produced by activated T cells, mast cells and basophils. IL-4 elicits many different biological responses, but has two dominant functions. The first is regulating differentiation of naive CD4+ T cell to the TH2 type. TH2 cells produce IL-4, IL-5, IL-10 and IL-13, which tend to favor a humoral immune response while suppressing a cell-mediated immune response controlled bu TH1 cells. The second is regulating IgE and IgG1 production by B cells. Recombinant IL-4 lacks all N-linked oligosaccharides to molecules highly overglycosylated, but all are fully biologically active. IL-4ís biological activity is species-specific.

Recombinant Human IL-4 produced in E. coli is a single, non-glycosylated polypeptide chain containing 130 amino acids and having a molecular mass of 15000D.
Biological ActivityHuman IL-4 is fully biologically active when compared to standards. The ED50 as determined by the dose-dependent stimulation of TF-1 cells is 0.2 ng/ml, corresponding to a specific activity of 5x10e6 units/mg.
Storage and stabilityLyophilized powder may be stored at 4°C for short-term only. Reconstitute to nominal volume by adding sterile ddH2O, 0.1% HSA or BSA, aliquot and store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
Amino acid sequenceThe sequence of the first five N-terminal amino acids was determined and was found to be Met-His-Lys-Cys-Asp.
Dimers and aggregates 1% as determined by silver-stained SDS-PAGE gel analysis.
Biological ActivityRecombinant Human Interleukin-4 is fully biologically active when compared to standard. The ED50 as determined by the dose-dependent stimulation of TF-1 cells is less then 0.2ng/ml, corresponding to a Specific Activity of 5x10 IU/mg.
ProteinProtein quantitation was carried out by two independent methods:
1. UV spectroscopy at 280nm using the absorbency value of 0.594 as the extinction coefficient for a 0.1% (1mg/ml) solution. This value is calculated by the PC GENE computer analysis program of protein sequences (IntelliGenetics).
2. Analysis by RP-HPLC, using a standard solution of IL-4 as a Reference Standard.
