Alpha-Internexin is a Class IV intermediate filament subunit expressed very early in neuronal development. It is related to but distinct from the better known neurofilament triplet proteins, NF-L, NF-M and NF-H, having similar protein sequence motifs and a similar intron organization. Many classes of mature neurons contain a-internexin in addition to NF-L, NF-M and NF-H. In some mature neurons a-internexin is the only neurofilament subunit expressed. Antibodies to internexin are therefore unique probes to study and classify neuronal types and follow their processes in sections and in tissue culture. In addition the very early developmental expression of a-internexin means its presence is an early and convenient diagnostic feature of neuronal progenitors cells and other cell committed to the neuronal lineage. The use of antibodies to this protein in the study of brain tumors has not been examined to date, but is likely to be of interest.

ApplicationsSuitable for use in Immunofluorescence, Western Blotting, Immunocytochemistry. Other applications not tested.
Recommended DilutionWestern Blot: 1:100. A 66kD band can be seen.
Immunocytochemistry: 1:25~1:50
Immunohistochemistry: Formalin-fixed cells in tissue culture, cryostat sections. Resistant to aldehyde fixation, so that this antibody is ideal for studies of formalin-fixed paraffin-embedded histological sections.
Optimal dilutions to be determined by the researcher.
Storage and StabilityMay be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Freeze at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
