| IL-17D, also called IL-27, is one of six IL-17 family members. All are homodimers with conserved cysteines that form a cystine knot structure. IL-17D (Accession # AF479775) is a 52 kD dimer expressed by resting CD4 + T cells and CD19 + B cells that acts on endothelial cells to stimulate the release of IL-6, IL-8, and GM-CSF. With other IL-17 family members, IL-17D recruits neutrophils to sites of inflammation. |

| Recommended Dilution | Western Blot: 1-2ug/ml. The detection limit is ~25ng/lane under non-reducing conditions. Reducing conditions are not recommended. |
| Flow Cytometry: 25ug/ml, 10ul labels 1-5x10e5 cells in a total reaction volume < 200ul. For intracellular staining, cells must be fixed and permeabilized using PBS, 4% paraformaldehyde, 0.1% saponin. Incubate for 30 minutes and wash cells with 0.1% saponin prior to addition of a secondary developing reagent. Cells should be washed a final time in 0.1% saponin prior to flow cytometric analysis. |

| Immunohistochemistry: 8-25ug/ml with appropriate secondary reagents to detect IL-17D in paraffin-embedded normal heart tissue sections. |

| Storage and Stability | Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. |
