The insulin receptor mediates insulin action through the phosphorylation of substrate proteins. The major substrates of the insulin receptor kinase are the insulin receptor substrate (IRS) family of proteins, IRS-1, IRS-2, IRS-3, and IRS-4. IRS proteins contain more than 70 potential Ser/Thr phosphorylation sites for kinases like PKA (cAMP-dependent protein kinase), PKC, and MAPK. The phosphorylation of Ser/Thr residues of IRS proteins has a dual function, serving either for a positive or negative modulation of insulin signal transduction. IRS proteins are also major substrates of the IGF-IR. Together with Insulin receptor, IRS proteins play an important role in the mitogenic signaling of both receptors.

Storage and StabilityMay be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
