The insulin receptor substrate 1 (IRS1) is a major effector of insulin receptor action. Unlike other activated receptors (e.g., the EGF receptor and the PDGF receptor) which directly bind to multiple signalling proteins, the activated insulin receptor primarily binds only IRS1 and then phosphorylates it on multiple tyrosine residues. Phosphorylated IRS1 serves as the major agent of insulin receptor-mediated signal transduction by being bound by cytoplasmic signaling proteins containing SH2 domains. Numerous SH2-containing signaling proteins (e.g., Grb2, SH-PTP2/SYP, PI 3-kinase, and Nck) have been shown to preferentially bind to distinct tyrosine phosphorylation sites in IRS1. The specificity and coordination of insulin receptor signaling may result from competition of these signaling proteins for binding to IRS1.

ApplicationsSuitable for use in Western Blot and Immunoprecipitation. Other applications not tested.
Recommended DilutionWestern Blot: 0.5-2ug/ml detected IRS-1 in a 3T3/A31 RIPA cell lysate.
Immunoprecipitation: 4ug immunoprecipitated IRS-1 from 500ug of a 3T3/A31 RIPA lysate.
Optimal dilution determined by the researcher.
Included Positive Antigen Control for Western Blot F4209: 3T3/A31 cell lysate.
Storage and StabilityMay be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, add sterile glycerol (40-50%), aliquot and store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
