The Janus family tyrosine kinases, consisting of Jak1, Jak2, Jak3 and Tyk2, associate with a variety of cytokine receptors and are activated by ligand binding to these receptors (1). Upon ligand binding to cytokine receptors, Jaks phosphorylate themselves and their associated receptors, thereby providing multiple binding sites for signaling proteins containing SH2 or other phospho-tyrosine-binding domains, including Stats (2), Shc (3), insulin receptor substrates (4) and focal adhesion kinase (5). The tyrosine residues 1007/1008 of Jak2 in the putative activation loop are the homologous tyrosine residues 1054/1055 in Tyk2, which are important in the regulation of Tyk2 kinase activity (6).

Storage and StabilityMay be stored at 4°C for short-term only. For long-term storage, store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
