| Human immunoglobulins are glycoproteins composed of two disulfide-bonded heavy (H) chain subunits, each of which is linked by interchain disulfide bonds to a light (L) chain forming a tetramolecular complex. There are five classes of immunoglobulins, designated IgG, IgA, IgM, IgD and IgE, which are defined by differences in the constant region of H chains. L chains are divided into kappa or lambda classifications based on structural antigenic differences. All classes of immunoglobulins have been found on the cell surface of B lymphocytes where they function as antigen receptors to elicit antigen-dependent proliferation and secretion of antigen specific soluble circulating antibodies. |

| Flow Cytometry Protocol | 1. 5x10e5 ficoll prepared human peripheral white blood cells were preincubated ~5 minutes with 20ul of 250ug/ml human Ig after which they were incubated 45 minutes on ice with 80ml of K0080-06B at 10ug/ml. |
| 2. Cells were washed twice and incubated with 2º reagent Streptavidin/R-PE (S7973-88P) after which they were washed three times, fixed and analyzed using a BD FACS Calibur. |
| 3. Cells stained positive with a mean shift of 2.17 log10 fluorescent units when compared to a Mouse IgG1/Biotin negative control at a similar concentration. |
| 4. Binding was 99% blocked when pre incubated 10 minutes with 20ul of 0.5mg/ml anti-Kappa antibody (K0080-06). |
| Storage and Stability | May be stored at 4°C for short-term only. For long-term storage, store at -20°C. Aliquots are stable for at least 6 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. |
