Keratin 4, which belongs to the type B (II) (basic) subfamily of high molecular weight keratins and exists in combination wth cytokeratin 13. It is primarily found in the non-cornifying squamous epithelium e.g. superficial and intermediate epithelial cells of the esophagus, ectocervix, toungue, vagina, larynx, pharynx, epiglottis, anus and the superficial cells of the cornea. Keratin 4 is also present in the suprabasal cells of the urinary bladder transitional epithelium in single cells and cell groups of sweat glands, prostatic ducts and in cylindrical ciliated bronchial epithelial cells. Keratin 4 may be detected in squamous cell carcinomas derived from several non-cornified stratified epithelia.

ApplicationsSuitable for use in Immunofluorescence, Western Blot and Immunohistochemistry. Other applications not tested.
Recommended DilutionsImmunohistochemistry (Frozen): 1:40 diluted in PBS. Gives excellent results in immunoperoxidase and immunofluorescence tests.
Immunohistochemistry (Paraffin): 1:10 after TUF treatment.
Optimal dilutions to be determined by the researcher.
Storage and StabilityMay be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

ImmunogenCytokeratin preparation purified from human esophagus.
SpecificityRecognizes human Keratin 4 at ~59kD. Stains non-cornifying squamous epithelium, cells in certain ciliated pseudostratified epithelia and ductal epithelia of various exocrine glands. Does not crossreact with Vimentin, Desmin, GFAP or Neurofilament. Does not crossreact with other cytokeratins.
