Keratins 6 and 16 are exprssed in keratinocytes, which are undergoing rapid turnover in the suprabasal region. Keratin 16 is absent in normal breast tissue and in noninvasive breast carcinomas. Only 10% of the invasive breast carcinomas show diffuse or focal positivity. A relatively high concordance was found between the carcinomas immunostaining with the basal cell and the hyperproliferation related keratins, but not between these markers and the proliferation marker Ki-67. This supports the conclusion that basal cells in breast cancer may show extensive proliferation, and that absence of Ki-67 staining does not mean that tumor cells Are not proliferating.

Applications	Suitable for use in Immunofluorescence and Immunohistochemistry (frozen and formalin/paraffin). Other applications have not been tested.
Recommended Dilution	Immunohistochemistry: 1-2ug/ml
	Staining of formalin/paraffin tissues requires boiling tissue sections in 10mM citrate buffer, pH 6.0, for 10-20 min., followed by cooling at RT for 20 min.
	Optimal dilutions to be determined by the researcher.

Storage and Stability	May be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, add sterile glycerol (40-50%), aliquot and store at -20°C or colder. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
