| Cytokeratin 19 is a member of type I acidic subfamily of intermediate filaments. it is expressed in various different human tissues except in liver. Keratin 19 is not expressed in hepatocytes, therefore, antibody to keratin 19 is useful in the identification of liver metastasis. | | | Catalog # | K0199-23X | | Applications | Suitable for use in Immunofluorescence, Western Blot, Immunohistochemistry. Other applications not tested. | | Recommended Dilution | Immunohistochemistry (frozen and formalin/paraffin): 1-2ug/ml, Staining of formalin/paraffin tissues is enhanced by digestion of tissue sections with pepsin at 1mg/ml Tris-HCl, pH 2.0 for 10-15 min at RT; with trypsin at 1mg/ml Tris-HCl (or PBS), pH 7.4 for 10-15 min at RT); or by boiling tissue sections in 10mM citrate buffer, pH 6.0, for 10-20 min., followed by cooling at RT for 20 min. (Protease XXIV damages the epitope recognized by this antibody). | | Western Blot: 2-4ug/ml for 2 hrs at RT. | | Optimal dilutions to be determined by the researcher. | | Positive Control | LS174T and HT29 cells, skin | | Cellular Localization | Cytoplasmic. | | Storage and Stability | May be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Freeze at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. | | CAS Number | n/a | | Clone Type | Monoclonal | | Isotype | IgG2a,k | | Clone No | 0.N.365 | | Host | Mouse | | Source | Human | | Concentration | ~1mg/ml | | Form | Supplied as a liquid in 10mM PBS, pH 7.4. Also available with BSA and azide. See K0199-23. | | Purity | Purified by Protein A affinity chromatography. | | Immunogen | Human breast cancer MCF-7 cells. Epitope: aa312-335 | | Specificity | Recognizes the rod domain of keratin 19 (40kD). Reacts with the MCF-7 cells, which are known to contain keratin 19. It shows no reaction with the cells lacking keratin 19 such as A431 and HeLa. Species Crossreactivity: human. Weak reactivity with rat, mouse and guinea pig. | | | Important Note | This product as supplied is intended for research use only, not for use in human, therapeutic or diagnostic applications without the expressed written authorization of United States Biological. |
