| Twenty human keratins are resolved with two-dimensional gel electrophoresis into acidic (pI <5.7) and basic (pI >6.0) subfamilies. The acidic keratins have molecular weights of 56.5, 55, 51, 50, 50’, 48, 46, 45, and 40kD. The basic keratins have molecular weights of 65-67, 64, 59, 58, 56 and 52kD. Members of acidic and basic subfamilies are found together in pairs. The composition of keratin pairs varies with cell type, differentiation status and environment. Many studies have shown the usefulness of keratins as markers in cancer research and tumor diagnosis. | | | Catalog # | K0199-90 | | Applications | Suitable for use in Immunofluorescence and Immunohistology . Other applications have not been tested. | | Recommended Dilution | Immunohistology (formalin/paraffin): 1-2ug/ml for 30 minutes at room temperature. | | Note: For staining of formalin-fixed tissues, digest sections with trypsin at 1mg/ml PBS, 10minutes at 37°C. | | Optimal dilutions to be determined by the researcher. | | Positive Control | Skin and lung carcinoma. | | Storage and Stability | May be stored at 4°C for short-term only. For long-term storage, store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. | | CAS Number | n/a | | Clone Type | Monoclonal | | Isotype | IgG1 | | Clone No | 0.N.368 + 0.N.370 | | Host | Mouse | | Source | Human | | Concentration | ~0.1mg/ml | | Form | Supplied as a liquid in PBS, pH 7.4, 0.2% BSA, 0.09% sodium azide, before the addition of 40% glycerol. Available without BSA and azide. See K0199-90X. | | Purity | Purified by Protein G affinity chromatography. | | Immunogen | Human epidermal keratin. Cellular Localization: Cytoplasmic. | | Specificity | Recognizes acidic keratins at 56.5, 50 (doublet), 48 and 40kD. Recognizes basic keratins at 65-67, 64, 59, 58, 56 and 52kD. Reacts with keratinized (56.5/65-67) and corneal (55/64) epidermis, stratified squamous epithelia of internal organs (51/59), stratified epithelia (50/58), hyperproliferative keratinocytes (48/56) and simple epithelia (45/52 and 46/54). Anti pan-keratin antibody cocktail differentiates epithelial from non-epithelial tumors. Species Crossreactivity: Human, monkey, bovine, rabbit, mouse, rat and chicken. | | | Important Note | This product as supplied is intended for research use only, not for use in human, therapeutic or diagnostic applications without the expressed written authorization of United States Biological. |
