Kinesin belongs to the group of motor proteins known to convert chemical energy released from ATP into mechanical energy. Conventional kinesin, member of the kinesin superfamily comprising more than 100 proteins, is involved in the anterograde vesicle transport inneuronal cells. Kinesin purified from mammalian brain homogenates is a heterotetramer consisting of two heavy (120-130kD) and two light chains (60-70kD), resulting in a molecular mass about 400kD. Each heavy chain contains an N-terminal globular motordomain with both a microtubule-binding site and an ATPase active center, stalk region responsible for heavy chain dimerization and finally C-terminal globular tail domain, which is implicated in cargo binding. Light chains may have a regulatory function.

Applications
Suitable for use in ELISA, Immunohistochemistry, Immunocytochemistry, Western Blot, Immunoprecipitation and RIA. Other applications not tested.
Recommended Dilutions
Immunohistochemistry: Paraformaldehyde fixed tissue. Also works for EM. Western Blot: 1:500 detected Kinesin (Heavy Chain) on 10ug mouse brain lysates. Mouse brain lystaes were resolved by electrophoresis, transferred to PVDF membrane and probed with K1780-01 (1:500). Proteins were visualized using an IgG (HRP) goat anti-mouse secondary antibody and a chemiluminescence detection system. Optimal dilution determined by the researcher.
Positive Control
Brain tissue
Storage and Stability
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Immunogen
Bovine brain kinesin
Form
Supplied as a liquid in PBS, pH 7.6, 0.09% sodium azide.
Purity
Purified by Protein A affinity chromatography.
Specificity
Recognizes bovine heavy chain of kinesin near the N-terminus. |Species Crossreactivity: All mammals, chicken, squid, sea urchins and clams.
