Clone Host Grade Applications
Polyclonal Rabbit Affinity Purified B
The Wnt family of secreted glycoproteins mediate cell-cell interaction during cell growth and differentiation in both embryos and adults.1,2 Canonical Wnt signaling by way of theb-catenin pathway is transduced by two receptor families, frizzled proteins and lipoprotein-receptor-related proteins 5 and 6 (LRP5/6) which binds Wnts and transmit their signal by stabilizing intracellular b-catenin.3-6 Kremen1 and Kremen2 are high-affinity Dkk1 receptors that functionally cooperate with the secreted protein Dickkopf1 (Dkk1) to block Wnt/b-catenin signaling.7 Kremen1 and Kremen2 are components of a membrane complex modulating canonical Wnt signaling through LRP6 in vertebrates.7 Dkk1 has been shown to inhibit Wnt signaling by binding to and antagonizing LRP5/6.8-10.

ApplicationsSuitable for use in Western Blot. Other applications not tested.
Recommended DilutionWestern Blot: 0.1-2ug/ml. Detects band at ~53kD.
Optimal dilutions to be determined by the researcher.
Storage and StabilityMay be stored at 4°C for short-term only. For long-term storage, store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
Clone TypePolyclonal
FormSupplied as a liquid in PBS, pH 7.4, 0.1% sodium azide, before the addition of glycerol to 40%.
PurityPurified by immunoaffinity chromatography.
ImmunogenSynthetic peptide derived from the C-terminal region of human, mouse and rat Kremen1.
SpecificityRecognizes Kremen1. Does not crossreact with the related Kremen2. Reactivity has been confirmed with myc-tagged mouse Kremen1-transfected 293T cell lysates. Species Crossreactivity: Human, mouse and rat. Based upon 100% amino acid conservation at this sequence, crossreactivity with human and rat is expected.
