Eukaryotic cells contain an abundant nuclear protein, Ku, which was initially discovered as an autoantigen. Ku is a heterodimer of 70 and 80kD subunits which binds to DNA ends and other alterations in double-stranded DNA including hairpins, nicks and gaps. Ku plays a role in DNA repair as an essential subunit of DNA-dependent protein kinase (DNA-PK) that phosphorylates certain transcription factors (Sp1, Oct-1 and p53). Ku targets DNA-PK to DNA. The Ku/DNA-PK complex might provide a structural framework for holding broken chromosomes together during repair. Ku plays a role in many diverse processes including cell signaling, proliferation, DNA repair, replication, transcriptional activation and apoptosis (2-4).
