Laminins are basement membrane components thought to mediate the attachment, migration and organization of cells into tissues during embryonic development by interacting with other extracellular matrix components. Laminin 5 is a complex glycoprotein composed of three subunits (alpha, beta, and gamma). Laminin 5 is thought to be involved in cell adhesion, signal transduction and differentiation of keratinocytes. Mutations in this gene have been identified as the cause of Herlitz type junctional epidermolysis bullosa. Alternatively spliced transcript variants encoding different isoforms have been identified.

ApplicationsSuitable for use in ELISA, Western Blot, Immunohistochemistry and Inhibition. Other applications not tested.
Recommended DilutionsWestern Blot: 1:500-1:2000 detects Laminin 5 in 10ug of human recombinant Laminin. A previous lot detected Laminin 5, gamma 2 chain in 10ug of Human Placenta lysates.
Immunohistochemistry (Paraffin): 5-10ug/ml; Do not over fix. A light 4% or 2% PFA is best. 4mm thick paraffin sections are dewaxed, rehydrated and immersed in 0.3% hydrogen peroxide-containing methanol for inactivation of intrinsic peroxidase, followed by treatment with Protease XXIV for 15 minutes at RT. Incubate at 37°C for 1 hour, followed by secondary antibody detection and visualize with DAB.
Inhibition: 40ug/ml inhibits a2b1 integrin binding to laminin 5. Optimal dilutions to be determined by the researcher.
Recommended ControlsHuman pancreatic tumor tissue, human breast carcinoma tissue, human placenta lysates, A431 whole cell lysates.
Storage and StabilityMay be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

ImmunogenGST fusion protein expressing aa382-608 of human laminin gamma 2 chain (III domain) (NP_005553).
SpecificityRecognizes the 150kD (precursor form) and 105kD (mature form) chain proteins of human Laminin 5. No reactivity is expected with other laminin types, as the gamma 2 chain of laminin is unique to Laminin 5. Strong positive staining is seen in basement membrane of the following tissues: skin, esophagus, lung and kidney. Positive staining is also seen in stomach, small intestine, spleen, thymus, prostate and ovary. Thyroid, testis, skeletal and cardiac muscle were negative for Laminin 5 staining.
