Laminins are large heterotrimeric, noncollagenous glycoproteins composed of alpha, beta, and gamma chains. Epithelial sheets in vivo are separated from the mesenchymal elements of the stroma by a thin layer of a specialized type of extracellular matrix termed the basement membrane (BM). This structure consists of individual components, some of which are ubiquitous in BMs and some are not. The ubiquitous ones comprise laminin (LN), entactin/nidogen (EN), collagen type IV (CIV), and large heparan sulfate proteoglycan (HSPG), which interact specifically with each other to form a continuous and regular BM. Alterations of BM integrity, from local discontinuities up to complete loss, are described in many types of human and animal epithelial neoplasms.

ApplicationsSuitable for use in Immunofluorescence, Immunoprecipitation, Western Blot and Immunohistochemistry. Other applications not tested.
Recommended DilutionImmunoprecipitation (Native and denatured): 2ug antibody/mg protein lysate using Protein G.
Western Blot: 1-2ug/ml for 2 hrs at RT.
Immunohistochemistry: 2ug/ml for 30 min at RT (Acetone-fixed frozen and alcohol-fixed, paraffin-embedded).
Optimal dilutions to be determined by the researcher.
Positive ControlLS174T cells. Normal colon or colon carcinoma.
Cellular Localization Basement membrane.
Storage and StabilityMay be stored at 4°C for short-term only. For long-term storage, store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
