The adaptor molecule LAT (linker for activation of T cells) is a palmitoylated integral membrane protein of 36-38kD, localized to the glycolipid-enriched microdomains in the plasma membrane and plays an important role in linking engagement of the TCR to the biochemical events of T cell activation. Upon TCR engagement, LAT becomes phosphorylated on multiple tyrosine residues by ZAP-70 and Syk PTKs and binds Grb2, PLC-gamma1, the p85 subunit of PI3K, and other critical signaling molecules, thereby recruiting these molecules to the plasma membrane. It is required for TCR-mediated Ca2+ mobilization and optimal tyrosine phosphorylation of phospholipase C-gamma1, Vav and SLP-76. It is also required for Erk activation, CD69 up-regulation, and AP- and NFAT-mediated gene transcription. The binding of these proteins to LAT appears to be critical to TCR-mediated signaling. It is usually expressed in thymus, T-cells, NK cells, mast cells and, at lower levels, in spleen. LAT usually exists in two known isoforms.
