Lipocalin 2 is a member of the lipocalin family of proteins which bind and transport small lipophilic molecules. Lipocalin 2 is released by activated neutrophils and occurs as 25kD glycosylated single protein chain monomers, which form dimers and small amounts of higher oligomers, as well as complexes with matrix metalloproteinase 9 (MMP-9; gelatinase B) (1). Low level expression of Lipocalin 2 in a variety of epithelia may be increased in inflammation or cancers (2). Expression of Lipocalin 2 in the kidney is dramatically increased by acute kidney injury (3).

ApplicationsSuitable for use in ELISA, Western Blot and Immunohistochemistry. Other applications not tested.
Recommended DilutionsELISA: Strong reaction seen when used as detection antibody in sandwich ELISA in combination with a polyclonal Lipocalin 2 capture antibody and when tested on recombinant Lipocalin 2 coated directly onto the well.
Western Blot: 1:50. Reacts with Lipocalin 2 in both reduced and unreduced forms in Western blotting of SDS-PAGE gels.
Immunohistochemistry: Reacts with Lipocalin 2 when staining paraformaldehyde-fixed sections containing neutrophils.
Optimal dilutions to be determined by the researcher.
Culture MediumRPMI 1640 with 10% fetal calf serum.
HybridomaX63-Ag8.653 myeloma cells with spleen cells from Balb/c mice.
Storage and StabilityMay be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
