| Protein acetylation of lysine, like phosphorylation of serine, threonine or tyrosine, is an important reversible modification controlling protein activity. The conserved amino-terminal domains of the four core histones (H2A, H2B, H3 and H4) contain lysines that are acetylated by histone acetyltransferases (HATs) and deacetylated by histone deacetylases (HDACs) (1). Acetylation of histones and transcription factors regulates | | | chromatin structure and gene activity (2-4). | | | Catalog # | L9058-02 | | Applications | Suitable for use in Western Blot and ELISA. Other applications not tested. | | Recommended Dilution | Western Blot: 1:1000, incubate membrane with diluted antibody in 5% BSA, 1X TBS, 0.1% Tween-20 at 4°C with gentle shaking, overnight. | | Peptide ELISA: 1:1000 | | Optimal dilutions to be determined by the researcher. | | Storage and Stability | May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. | | CAS Number | n/a | | Clone Type | Monoclonal | | Isotype | IgG2a | | Clone No | 3A49 | | Host | Mouse | | Concentration | Not determined | | Form | Supplied as a liquid in 10mM HEPES, pH 7.5, 150mM sodium chloride, 0.1mg/ml BSA, 50% glycerol. | | Purity | Ascites | | Immunogen | Synthetic acetylated lysine-containing peptide (KLH) | | Specificity | Recognizes Acetylated-Lysine proteins only when post-translationally modified by acetylation on the epsilon-amine groups of lysine residues. Detection of acetylated lysine by this antibody is largely independent of the surrounding amino acid sequence. The antibody has been shown to recognize acetylated proteins including histones, p53, CBP, PCAF and chemically acetylated BSA. Species Crossreactivity: A broad species crossreactivity is expected based on conservation of sequence homology. | | | Important Note | This product as supplied is intended for research use only, not for use in human, therapeutic or diagnostic applications without the expressed written authorization of United States Biological. |
