Other applications not tested. | | Recommended Dilution | Western Blot: 0.5-2ug/ml detects acetylated histones in acid-extracted proteins from sodium butyrate (5mM) treated HeLa cells. Sodium butyrate, an inhibitor of deacetylases, was used to increase the level of acetylated histones. 0.5-4ug/ml detected: chemically acetylated BSA (10ng), GST-p53 (400ng) acetylated in vitro with either recombinant p300 or PCAF, auto-acetylated p300 (400ng) and PCAF (400ng). Proteins were resolved by electrophoresis, transferred to nitrocellulose and probed with 4ug/ml L9058-02A. Proteins were visualized using a goat anti-mouse secondary antibody conjugated to HRP and a chemiluminescence detection system. | | Immunoprecipitation | 5ug immunoprecipitates in vitro acetylated PCAF added to a 3T3 RIPA cell lysate. The immunoprecipitated PCAF was detected by subsequent immunoblot analysis using 1ug/ml monoclonal anti-GST. | | Optimal dilutions to be determined by the researcher. | | Storage and Stability | May be stored at 4°C for short-term only.
