| Maltose binding protein is a bacterial protein, which is often used in protein expression studies because it creates a stable fusion product that does not appear to interfere with the bioactivity of the protein of interest. It also allows for its easy purification from bacterial extracts under mild conditions. Protein tagging has become a widely applied method for ready purification of proteins. The specific interaction between the tag and a solid-phase ligand allows for the selective binding and elution of the tagged protein. Many vectors that incorporate these tags have been engineered with a cleavage site between the tag and the protein of interest. This permits the separation of the protein from the tag, if needed. One commonly used system is the maltose-binding protein (MBP) (1). In this system, the target gene is inserted in the appropriate vector and expressed.
