MAP kinase kinase 6 (Mek6/MKK6) is a 38 kD dual-specificity protein kinase that is abundant in skeletal muscle and is activated by UV light, anisomycin, osmotic shock and inflammatory cytokines, but not by phorbol esters, NGF or EGF (1). Mek6 phosphorylates and activates MAP kinases such as p38 (2,3) and p38 beta (4). Mek6 is phosphorylated and activated by ASK1, a MAPKKK that induces apoptosis when overexpressed (5). Mek6 also interacts with mixed lineage kinase-3 (MLK-3), a kinase that participates in stress response kinase cascade, possibly by phosphorylating MKK4, an isoform of Mek6 (6).
