Mast cells contain a number of preformed chemical mediators such as histamine, chymase, carboxypeptidase and proteolytic tryptase. A substantial quantity of chymase is estimated to be found in mast cells of skin and lung and suggests this enzyme plays a major role in mast cell mediated events. Human mast cell chymase is considered to be an important marker of mast cells as well as an important mediator of inflammation.

ApplicationsSuitable for use in Western Blot and Immunohistochemistry. Other applications have not been tested.
Recommended DilutionImmunohistochemistry (Formalin/paraffin): 1:2000 for 20min at RT. Staining of formalin-fixed tissues requires boiling tissue sections in 10mM citrate buffer, pH 8.0, for 10-20min followed by cooling at RT for 20 min.
Optimal dilutions to be determined by the researcher.

Storage and StabilityMay be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

SpecificityRecognizes human mast cells in routinely fixed tissues, and has been used to study the distribution of mast cells in skin, synovium, lung and heart. Binding of this antibody to chymase is not reduced in the presence of heparin, suggesting that the antibody does not bind to the heparin-binding domain of chymase
