Mast cells contain a number of preformed chemical mediators such as histamine, chymase, carboxypeptidase and proteolytic tryptase. A substantial quantity of tryptase is estimated to be found in mast cells of skin and lung and suggestes this enzyme plays a major role in mast cell mediated events. In vitro studies indicate tryptase can cleave C3 to form C3a anaphylatoxin, inactivate fibrinogen as a coaguable substrate for thrombin and activate latent collagenase. Human mast cell tryptase has considerable potential as a marker for mast cell activation as well as being a mediator of inflammation.

ApplicationsSuitable for use in Western Blot and Immunohistochemistry. Other applications not tested.
Recommended DilutionImmunohistochemistry: 1:100-1:500. Tissues or cytospin preparations should be fixed in Carnoy’s Fluid (60% ethanol, 30% chloroform, 10% glacial acetic acid). Not recommended for use on formaldehyde fixed tissue. May be used on thin sections prepared for ultrastructural analysis after fixation of tissue in a modified Karnovsky’s reagent.
Optimal dilutions to be determined by the researcher.
Storage and StabilityMay be stored at 4°C before opening. DO NOT FREEZE! Stable at 4°C as an undiluted liquid. Dilute only prior to immediate use. Stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. Freezing alkaline phosphatase conjugates will result in a substantial loss of enzymatic activity.
