| MGMT is a p53 related DNA repair protein that protects tissues against toxic and carcinogenic effects of alkylating agents and thus remains the first line defense against alkylation damage of DNA. This protein functions by a unique stoichiometric and suicidal reaction mechanism in which the alkyl groups bound to the O6-position of guanine are transferred to a cysteine in its active site, resulting in the direct restoration of the normal base and self-inactivation of MGMT protein. It is highly expressed in human cancers and is also a central determinant of tumor resistance to many clinically used anticancer alkylating agents.
