| O6-methylguanine-DNA transferase (MGMT) is a ubiquitous 22kD DNA repair protein, involved in the repair of mutagenic, carcinogenic, and cytotoxic O6-alkylguanine adducts, including primary alkyl adducts, induced by the chloroethylating and methylating agents used in the treatment of cancers. | of MGMT molecules within the cell and the rate of MGMT synthesis. MGMT repair thus represents a major form of chemotherapeutic drug resistance. The importance of MGMT in determining clinical response, both to chloroethylnitrosourea chemotherapeutic agents and to carcinogenesis by methylating agents, has been described. MGMT expression is highly variable in normal tissues as well as in cancer cells.10 A fraction of primary tumor cells and ~20% of human tumor cell lines lack MGMT expression. Hypermethylation of the Mgmt promoter region has been linked to loss of MGMT protein expression in cancer cell lines and tissues. Tumor cell lines lacking MGMT are highly sensitive to alkylating agents and nitrosourea-type agents, suggesting that selective inhibition of MGMT in tumor cells may be an effective strategy in overcoming chemotherapeutic drug resistance. |

| Storage and Stability | May be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, add sterile 40-50% glycerol, aliquot and store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. |
